You might want to check out FreeDOS
http://www.freedos.org/
It is an open source MS DOS compatible OS that is still under active
development. If you want 32 bit DOS you can probably build it from the
source.
FreeDOS aims to be a complete, free, 100% MS-DOS compatible operating
system (mostly achieved except Windows compatibility - Windows
standard-mode works on FreeDOS, but 386-mode / WfW 3.11 does not.)

Sounds good but I don't see any mention of USB support.

USB is essentially a serial port and serial ports are supported. I'll
contact them to ask about USB support and let you know what I find out.
