So home I tested an OTG cable,

Rooted Galaxy S4: Read and Write to usb thumbdrive using ES File Explorer

I also plugged a usb mouse into the Note 5 and a functioning pointer appeared on the screen without having to do anything besides just plugging it in.

The main disadvantage to Rooting your device is that it voids your warranty.
You can think of rooting your phone much like giving yourself SU rights in linux. It lets you do more, but it also lets you screw stuff up if you are not careful.
It appears that root is probably not necessary for using usb otg, though I had read some comments it was needed for thumbdrives and not devices like mice and game controllers.
